特异性 DNA-PKcs Rabbit Recombinant mAb detects endogenous levels of total DNA-PKcs.
背景 DNA-PKcs is a catalytic subunit of DNA-PK, a nuclear DNA-dependent serine/threonine protein kinase. DNA-PK is comprised of three key components (Ku70/80 and DNA-PKcs) and is a member of the phosphatidylinositol-3-kinase (PI3K) family. DNA-PK is involved in V(D)J recombination and also strongly implicated in telomere maintenance. Like Ku, DNA-PKcs is located at telomeres and has a role in telomere capping, however, it does not affect telomere length or telomerase activity. DNA-PKcs protects telomeric DNA and chromosome ends. DNA-PK activation and post-translational modifications are numerous and complex. The autophosphorylation of DNA-PK results in kinase inactivation and dissociation of DNA-PKcs from the heterodimer Ku. In addition, Ku (as the DNA recognition component) and DNA-PKcs could influence transcription. DNA-PK (comprising both DNA-PKcs and Ku) functions as a potent inhibitor of transcription initiation by Pol I and that this inhibition involves ATP hydrolysis and Pol I phosphorylation. In addition to RNA-Pol I, DNA-PK has also been shown to phosphorylate RNA Polymerase II (RNA-Pol II) and a variety of sequence specific regulatory transcription factors. DNA-PKcs may act as a scaffold protein to aid the localization of DNA repair proteins to the site of damage. It is autophosphorylated after cellular IR exposures, which modifies its binding with Ku and has been implicated in the phosphorylation of a wide range of DNA damage/checkpoint proteins. This auto-phosphorylation event is critical for correct NHEJ activity within the cell.
