背景 DNA methylation is involved in epigenetic regulation of gene expression, in the context of X chromosome inactivation, genomic imprinting, and development. In the cell, Dnmt1 is the major enzyme in maintenance of the pattern of DNA methylation after DNA replication. The protein is located at the replication fork, where it could directly modify nascent DNA immediately after replication. Dnmt1 prefers hemimethylated DNA over unmethylated DNA up to 40-fold in vitro. DNMT1 is highly expressed during the S phase and mostly localized to nuclei of somatic cells during G1 and G2 phases and is actively targeted to replication foci during the S-phase. DNMT1 is found to be either downregulated or upregulated during cellular differentiation. In many instances, for example in muscle cells, F9 mouse tumor cells, PC12 neuronal cells and in skin cells DNMT1 expression and/or activity levels are decreased in cells when differentiation is induced experimentally. DNMT1 expression is upregulated in certain malignant blood cells. DNMT1 is also shown to contribute to the cellular response to DNA damage. DNMT1 is recruited rapidly and transiently to DNA double strand breaks and interacts with multiple components of the DNA damage response machinery including PCNA and the ATR effector kinase CHK1. DNMT is highly expressed in cancer cells and associated with cellular transformation.
