背景 | Glutamate clearance is facilitated by a family of Na+- and K+-coupled excitatory amino acid transporters (EAATs). Five mammalian EAATs have been cloned and characterized: EAAT1 (known as GLAST in rodents), EAAT2 (known as GLT-1 in rodents), Excitatory amino acid transporter 2 (EAAT2), EAAT3 (known as EAAC1 in rodents), EAAT4 and EAAT5. In the CNS, while EAAT1 and EAAT2 are found primarily in astrocytes, EAAT3 and EAAT4 are found principally in neurons. EAAT5 is expressed primarily in the retina. The glial glutamate transporters, EAAT1 and EAAT2, are found primarily on perisynaptic processes of astrocytes closely associated with excitatory synaptic contacts and are responsible for maintaining low extracellular glutamate concentrations. This is particularly the case for EAAT2, which may be responsible for up to 80–90% of all extracellular glutamate uptake activity. Dysfunction or reduced expression of EAAT2 has been documented in both chronic and acute neurodegenerative disorders. |
