背景 Control of mRNA translation plays a critical role in cell growth, proliferation and differentiation. In eukaryotes, most mRNAs are translated in a cap-dependent manner. The cap structure m7GpppN (where N is any nucleotide) is found at the 5' terminus of all cellular eukaryotic mRNAs (except those in organelles). The mRNA 5' cap-binding protein eIF4E is a key player in the regulation of translation. eIF4E is regulated at multiple levels, including gene expression, sequestration and phosphorylation, etc. It mediates a rate-limiting process that drives selective translation of many oncongenic proteins such as cyclin D1, survivin and VEGF, thereby contributing to tumour growth, metastasis and therapy resistance.
