特异性 UBA1 Rabbit Recombinant mAb detects endogenous levels of total UBA1.
背景 The E1-type ubiquitin-activating enzyme (UBA1) is responsible for ubiquitin activation, the initial step of ubiquitin-protein ligation. UBA1 activates ubiquitin through a two-step intramolecular, ATP-dependent reaction to generate a high energy E1-thiol ester-ubiquitin intermediate; the activated ubiquitin moiety is then transferred onto ubiquitin-conjugated enzyme E2, serving as a carrier of activated ubiquitin. Inactivation of UBA1 gene is lethal in yeast, causing a deficiency in protein turnover by ubiquitin-proteasome pathway. During G1 and G2 phase, UBA1 is almost exclusively nuclear, whereas in other mitotic phases it is present in both the nucleus and the cytoplasm. UBA1 can be phosphorylated at several serine residues, again closely linked to cell cycle status; UBA1 phosphorylation is maximal during G2 phase. UBA1 is also implicated in DNA repair pathways: UBA1 is essential for protein ubiquitylation-mediated repair of double-strand DNA breaks, playing a critical role in regulating protein homeostasis.
