特异性 Wnt5a Rabbit Recombinant mAb detects endogenous levels of total Wnt5a.
背景 Wnt5a is a noncanonical Wnt ligand that is evolutionarily conserved and plays an important role in development. It can inhibit or activate canonical Wnt/β-catenin signaling pathway in a receptor context-dependent manner. Wnt5a signaling is critical for regulating normal developmental processes, including stem cell self-renewal, proliferation, differentiation, migration, adhesion, and polarity. Wnt5a regulates cellular functions by binding to several Fzs. Among the mammalian Fz family, Wnt5a signalling could be mediated through at least Fz3, Fz4, Fz5 and Fz8 but not Fz6 from the assessment of the phosphorylation of dishevelled (Dvl orthologue) in Drosophila S2 cells. Fz7 is required for Wnt5a-induced Dvl polymerization and AP-1 activation in mouse fibroblast L cells. Wnt5a and Wnt11 activate β-catenin-independent pathway, which primarily modulates cell movement, as initially observed during embryogenesis. Wnt5a is involved in PCP pathway (such as stereociliary bundle orientation in the cochlea) in mouse mediated by Fz and Dvl and activates small G proteins, including Rac and Rho, JNK, and Rho-associated kinase (Rho-kinase). Wnt5a has also been suggested to regulate the Ca2+ pathway. In mammalian cells Wnt5a elicits an intracellular release of Ca2+, thereby activating PKC, CaMKII, and calpain to regulate cellular functions including cell migration. In addition, Wnt5a regulates other pathways, such as those involving Wnt5a-Dvl-atypical PKC (aPKC), Wnt5a-Ror2-filamin and Wnt5a-Fz2-Dvl-adenomatous polyposis coli gene product (APC)-paxillin. The Wnt5a-Dvl-aPKC pathway promotes axon differentiation of hippocampal neurones.
