体外研究(In Vitro)
体外研究活性

在基于细胞的IPKA (C33A)试验中,Akti-1/2抑制Akt1和Akt2,IC50分别为305 nM和2086 nM。在HT29,MCF7,和A2780细胞中,Akti-1/2通过急剧增加caspase-3活性诱导细胞凋亡。[1]

在肝细胞中,Akti-1/2阻断胰岛素调节PEPCK,G6Pase表达,和FOXO1活性。[2]

Akti-1/2也会通过阻断PKB强烈增强PAR-1-介导的血小板聚集。[3]

在HCC827,NCI-H522,NCI-1651,和PC-9细胞中,Akti-1/2抑制细胞生长,IC50 为4.7 μM,7.25 μM,和9.5 μM,结合gefitinib时,Akti-1/2会增强细胞生长和凋亡的抑制。[4]

激酶实验 激酶筛选
简而言之,所有试验(25.5 μl在21°C下进行30 min)使用Biomek 2000实验室自动化工作站在96孔格式中进行。反应包含5–20 mU纯化的激酶和底物肽或蛋白质,通过加入10 mM 乙酸镁和5,20,或50 μM ATP ([γ-33P]-ATP, 800 cpm/pmol)起始。
