背景 | NF-κB is a ubiquitous heterodimeric transcription factor prototypically composed of p50 and p65 polypeptides related to the v-Re1 oncoprotein. NF-KB mediates the regulated expression of multiple immunomodulatory genes bearing cis-acting κB enhancer elements, including the K light chain of Igs, leukocyte adhesion molecules, class I MHC molecules and the 1L-2 receptor α-chain, and viral promoters including that of HIV. In unstimulated cells, the NF-κB dimers are sequestered in the cytoplasm by a family of inhibitors, called IκBs (Inhibitor of κB), which are proteins that contain multiple copies of a sequence called ankyrin repeats. The IκB proteins mask the nuclear localization signals (NLS) of NF-κB proteins and keep them sequestered in an inactive state in the cytoplasm. The IκB family consists of IκBα, IκBβ, IκBε, and Bcl-3, the best-studied and major IκB protein is IκBα. In most cells, activation and translocation of NF-KB are contingent upon release from IkBα. The prevailing model for NF-KB activation argued that IkBα becomes phosphorylated in response to a variety of environmental stimuli that cause it to dissociate from the NF-KB complex before its proteolysis. Whereas some reports indicate that NF-KB does not dissociate from IkBα following its phosphorylation and that more likely proteolysis of the phosphorylated IkBα will result in NF-KB release. The mutation of IkBa S32 and S36 abrogates the stimuli-induced phosphorylation and subsequent degradation. |
