背景 IL-2最初作为T细胞生长因子(TCGF)被发现,在抵御病原体感染的免疫反应中发挥着关键作用。TCR识别并结合外源抗原,刺激T细胞分泌IL-2并表达IL-2R。随后,IL-2/IL-2R相互作用激活细胞内Ras/Raf/MAPK、JAK/STAT和PI3K/AKT信号通路,最终刺激抗原选择的细胞毒性T细胞的生长、分化和存活。人IL-2可作用于小鼠和人T细胞,其受体可被其他细胞因子共享。IL-2Rα是IL-2特异性受体,IL-2Rβ与IL-15共享,而γ链是包括IL-2、IL-4、IL-7、IL-9、IL-15和IL-21在内的许多细胞因子的共同受体。

  • Human peripheral blood mononuclear cells (PBMC) were stimulated with PMA and calcium ionophore A23187 for 6 hours, with Golgi blocker added (final concentration 2 uM). Cells were first stained for surface markers with mouse anti-human CD3-FITC; then fixed, permeabilized, and stained for intracellular cytokine using AF647 Human IL-2 Antibody [MQ1-17H12] (Product No.: H1227). Analyzed data are derived from total cells.
