特异性 Apg12(Atg12) Rabbit Recombinant mAb detects endogenous level of total Apg12(Atg12).
背景 In eukaryotic cells, autophagy is the major degradative pathway by which proteins and organelles are delivered from the cytosol to lysosomes/vacuoles for breakdown and recycling. Triggering of autophagy results in the formation of double membrane-bound vesicles termed autophagosomes. The conserved Atg5–Atg12/Atg16 complex is essential for autophagosome formation. Atg12, a post-translational modifier, is activated and conjugated to Atg5 by a ubiquitin-like conjugation system. The carboxyl-terminal glycine of Atg12 is activated by Atg7, an E1-like enzyme, through ATP-hydrolysis. And Atg12 is then transferred to Atg10, an E2-like enzyme. Finally, Atg12 is conjugated to Atg5 and the Atg12-Atg5 conjugate forms a multimeric complex with Atg16. Atg16 is required for the localization of the Atg5-Atg12 conjugate to the PAS in yeast and isolation membranes in higher eukaryotes. The formation of Atg12-Atg5 is constitutive and is not regulated by starvation. The Atg12-Atg5 conjugate is essential for autophagy.
