背景 Calpain is a cytoplasmic cysteine protease requiring calcium ions for activity. It is implicated in a variety of calcium-regulated cellular processes such as signal transduction, cell proliferation, cell cycle progression, differentiation, apoptosis, membrane fusion, and platelet activation while deregulation of its activity has been implicated in various pathological conditions such as neuronal degeneration, Alzheimer’s disease, metastasis, and cataract. Calpain exists in the cytosol as an inactive enzyme and translocates to membranes in response to increases in the cellular Ca2+ level. At the membrane, calpain is activated in the presence of Ca2+ and phospholipids. Activated calpain hydrolyzes substrate proteins at membranes or in cytosol after release from membranes. Calpain 2 is regarded as a ubiquitous calpain. It is a heterodimer composed of a large catalytic subunit of 80 kDa and a common regulatory subunit of 30 kDa (Calpain4) that dissociates upon activation.
