背景 Citrate synthase is a component of nearly all living cells and plays a key role in the central metabolic pathway of aerobic organisms, the citric acid cycle. The citric acid cycle functions as a source of two reducing equivalents for the electron transport chain and as a source of intermediates required for the biosynthesis of amino acids for ketogenesis, lipogenesis, and gluconeogenesis. Citrate synthase catalyzes an important step within the cycle, the Claisen condensation of acetyl-Coenzyme A with oxaloacetate to form citrate; and it is the only enzyme in the cycle that can catalyze the formation of a carbon-carbon bond. Citrate acts not only as a substrate for the citric acid cycle within the mitochondria, but also as an acetyl donor for acetyl-Coenzyme A synthesis by ATP-citrate-Iyase outside the mitochondria after transport through the mitochondrial membrane.
