背景 Ionotropic glutamate receptor coassemble to form ligand-gated ion channels containing an agonist recognition site, a transmembrane ion permeation pathway, and gating elements that couple agonist-induced conformational changes to the opening or closing of the permeation pore. Glutamate receptors mediate fast excitatory synaptic transmission in the central nervous system and are localized on neuronal and non-neuronal cells. These receptors regulate a broad spectrum of processes in the brain, spinal cord, retina, and peripheral nervous system. The glutamate receptors assemble as tetrameric complexes of subunits, and functional receptors are formed exclusively by assembly of subunits within the same functional receptor class. Glutamate receptors are grouped into four distinct classes based on pharmacology and structural homology, including the AMPA receptors (GluA1-GluA4), the kainate receptors (GluK1-GluK5), the NMDA receptors (GluN1, GluN2A-GluN2D, GluN3A, and GluN3B), and the δ receptors (GluD1 and GluD2). This class of receptors has also been shown to be involved in many diverse neuropathologies, such as ischemia, Parkinson's disease, and amyotrophic lateral sclerosis, and to play other varying roles throughout the body.
