背景 | Glutamate receptor (GluRs) are important in neural plasticity, neural development, and neurodegeneration. Activation of GluRs mediate a large number of neuronal processes such as long-term potentiation and ischemic damage. Two classes of GluRs have been characterized: ionotropic (iGluRs) and metabotropic (mGluRs). They both bind to Glu, and Glu binding can influence the permeability of ion channels. The iGluRs are ligand-gated ion channels that mediate the vast majority of excitatory neurotransmission in the brain. The iGluR family is classified into three major subtypes: NMDA, AMPA and kainate (Ka) receptors. AMPA family is composed of GluR 1-4 (GluRA-D). AMPA receptors are involved in mediating most forms of fast glutamatergic neurotransmission, which corresponds to a Ca2+ influx. GluR1 has several identified phosphorylation sites on the intracellular carboxy terminus: serine (S) 818, S831, threonine (T) 840, and S845. |
