在HEK-293细胞中，GSK-J1抑制瞬时转染的JMJD3和UTX活性。GSK-J1通过增加总细胞核H3K27me3水平，也会抑制原代巨噬细胞产生TNF-α。

在MC3T3-E1细胞中，GSK-J1抑制Runx2和Osterix表达和ALP活性，并增加总体H3K27me3水平。

激酶实验 JmjD3 和 UTX活性的MALDI-TOF分析
纯化的JmjD3 (1 μM)和UTX (3 μM)与10 μM多肽[Biotin-KAPRKQLATKAARK(me3)SAPATGG]在50 mM HEPES pH 7.5，150 mM KCl，50μM (NH4)2SO4•FeSO4•H2O，1 mM 2-氧化戊二酸，和2 mM 抗坏血酸(JmjD3，25°C下3分钟；UTX，25°C下20分钟)中，在不同浓度抑制剂 (0，0.005，0.01，0.02，0.05，0.1 μM)存在下进行培养。加入10mM EDTA停止反应。反应通过ZipTip脱盐，并用α氰基-4-羟基肉桂酸MALDI基质在MALDI板上点样。样品在MALDI-TOF R系统中进行分析。
