特异性 Iba1 Rabbit Recombinant mAb detects endogenous level of total Iba1.
背景 Ionized calcium binding adaptor molecule 1 (Iba1) is a microglia/macrophage-specific calcium-binding protein. Iba1 has the actin-bundling activity and participates in membrane ruffling and phagocytosis in activated microglia. Iba1 forms complexes with L-fimbrin in membrane ruffles and phagocytic cups and co-operates with L-fimbrin in modulating actin reorganization to facilitate cell migration and phagocytosis by microglia. Expression of Iba1 is up-regulated in activated microglia following facial nerve axotomy, ischemia and several brain diseases. It is involved in the signalling pathways of calcium and a Rho family small GTPase, Rac, which is the essential molecule in regulating actin reorganization in membrane ruffling. Iba1 binds to phosphatidylserine in the presence of calcium, translocates to the phosphatidylserine-rich inner surface of the cell membrane in a calcium-dependent manner and also translocates to the cell membrane during membrane ruffling. Iba1 is also shown to bind to phosphatidylinositol 4,5-bisphosphate, the significant substrate of PLC-γ. Iba1 may, directly or indirectly, support translocation of PLC-γ to the cell membrane and its substrate, and may induce Rac activation by potentiating the activity of PLC-γ. Mutant Iba1 effectively suppresses the membrane ruffling produced by stimulation with macrophage colony-stimulating factor (M-CSF) or by expression of dominant active RacV12.
