背景 Protein deglycase DJ-1, also known as Parkinson disease protein 7 (PARK7) is a multifunctional protein that plays essential roles in tissues with higher order biological functions such as the testis and brain. DJ-1 has multiple functions: it works as an antioxidant, in transcriptional regulation, as a molecular chaperone and in protein degradation. DJ-1 is located in the cytoplasm, nucleus, and mitochondria in cells, and secreted DJ-1 has been observed in various cultured cells and tissues, including cancer cells and tissues. DJ-1 is translocated from the cytoplasm to nucleus upon exposure to growth factors, and oxidation of C106 described later is necessary for nuclear translocation of DJ-1 [61]. DJ-1 contains three cysteine residues, C46, C56, and C106. DJ-1 is also modified by sumoylation, S-nitrosylation, and phosphorylation.
