背景 磷酸化NF-κB p65(Ser536)是活化的RelA/p65的标志物,突显了增强NF-κB转录活性的磷酸化事件。NF-κB/Rel家族的转录因子是炎症和免疫反应的核心调节因子。哺乳动物家族成员包括RelA(p65)、c-Rel、RelB、NF-κB1(p105/p50)和NF-κB2(p100);p105和p100经蛋白酶体加工成p50和p52,它们与Rel蛋白二聚化以结合DNA并控制转录。在静息细胞中,NF-κB二聚体被IκB抑制蛋白隔离在细胞质中。活化刺激诱导IκB磷酸化,使其被泛素依赖性蛋白酶体降解,释放NF-κB易位至细胞核并调节基因表达。NIK和IKKα(IKK1)控制p100向p52的磷酸化和加工,使p52核易位。
