特异性 Phospho-AMPK alpha 2 (S491) Rabbit Recombinant mAb detects endogenous levels of phosphorylated AMPK alpha 2 (S491).
背景 AMPK is an evolutionarily conserved serine/threonine kinase that was originally identified as the key player in maintaining cellular energy homeostasis. AMPK regulates diverse metabolic and physiological processes and is dysregulated in major chronic diseases, such as obesity, inflammation, diabetes and cancer. AMPK is a heterotrimeric complex containing one catalytic α-subunit and two regulatory β- and γ-subunits. In mammals, AMPK α- and β-subunits have two isoforms each, and AMPK γ-subunit has three isoforms. In addition to phosphorylation on the critical T172 residue, a number of secondary phosphorylation sites on the α and β subunits have been identified. Phosphorylation at these sites may regulate AMPK activation. For example, α1 S485 and α2 S491 phosphorylation is correlated with decreased AMPK activity in cardiac tissue in response to insulin treatment. α2 S491 phosphorylation by p70S6K kinase, acting downstream of PI3K-AKT, is found to underlie AMPK inhibition in the hypothalamus.
