背景 Glycogen synthase kinase-3 (GSK-3) is a multifunctional serine/threonine kinase found in all eukaryotes. The enzyme is a key regulator of numerous signalling pathways, including cellular responses to Wnt, receptor tyrosine kinases and G protein-coupled receptors and is involved in a wide swathe of cellular processes ranging from glycogen metabolism to cell cycle regulation and proliferation. GSK-3 is unusual in that it is usually constitutively active in cells and is regulated through inhibition of its activity. The activities of GSK-3 are positively regulated by phosphorylation of Tyr279 and Tyr216 for the α- and β-isoforms, respectively. Phosphorylation of Tyr279 in GSK-3α and Tyr216 in GSK-3β has been reported to facilitate GSK-3 activity by inducing rotation of the tyrosine side chain, which moves out of the substrate-binding groove, promoting substrate accessibility.
