特异性 Phospho-STAT1 (S727) Rabbit Recombinant mAb detects endogenous levels of phosphorylated STAT1 (S727).
背景 Serine phosphorylation has generally been considered indispensable for full transcriptional activity of STAT proteins. STAT1 is a cytoplasmic protein that, upon awakening from dormancy by signaling from IFNs and other factors, activates numerous genes designed, in principle, to defend the host. STAT1 acts as a tumor promoter in leukemia. The activity of Stat1 is regulated by phosphorylation on Tyr701 by Janus kinases (JAKs), important for its dimerization, translocation to the nucleus, and binding to DNA. A second phosphorylation, of Ser727, is required for transcriptional responses to certain inducers, eg, IFN-γ, but not to others, eg, IFN-α. In accordance with this requirement, both Stat1 Tyr701 and Ser727 phosphorylation are necessary for growth arrest in response to IFN-γ, whereas IFN-α—induced growth arrest requires only Tyr701 phosphorylation.
