背景
TGF-β1(转化生长因子β1)是庞大TGF-β超家族中三个高度同源的哺乳动物成员之一,均具有特征性胱氨酸结结构。TGF-β1、-2和-3作为高度多效性细胞因子,扮演调控免疫功能、增殖和上皮-间质转化的细胞开关,各亚型功能不可互相替代;尤其值得注意的是,TGF-β1基因敲除小鼠会出现造血功能和内皮分化缺陷,最终死于过度炎症反应。人源TGF-β1 cDNA编码390个氨基酸的前体蛋白,包括29个氨基酸的信号肽和361个氨基酸的前体蛋白,经弗林蛋白酶样转化酶切割为N端249个氨基酸的潜伏相关肽(LAP)和C端112个氨基酸的成熟TGF-β1;二硫键连接的LAP同源二聚体在分泌后仍与成熟TGF-β1非共价结合形成小潜伏复合物,而LAP可与三种潜伏TGF-β结合蛋白(LTBP)之一共价连接,构成能与细胞外基质相互作用的大潜伏复合物。其活化途径涉及纤溶酶、基质金属蛋白酶、血小板反应蛋白1及特定整合素。成熟人源TGF-β1与猪、狗、牛直系同源物具有100%氨基酸一致性,与小鼠、大鼠、马同源物达99%,显示跨物种活性。信号传导始于与TGF-β RII(II型丝/苏氨酸激酶受体)的高亲和力结合,进而磷酸化并激活TGF-β RI(ALK-5或ALK-1),引发Smad蛋白磷酸化与转录调控,该过程受辅助受体β聚糖(TGF-β RIII)和内格林的调节,或通过Smad非依赖性通路实现环境依赖性效应。
