背景 Retinoids, the natural and synthetic vitamin A derivatives, are known to regulate a broad range of biological processes, including growth, differentiation, and development. The effects of retinoids are mainly mediated by two classes of nuclear receptors: the retinoic acid (RA) receptors (RARs) and the retinoid X receptors (RXRs). RARs and RXRs are members of the steroid-thyroid hormone receptor superfamily that also includes receptors for estrogen and vitamin D. Three RAR subtypes [RARα, RARβ, and RARγ] that originate from three distinct genes have been identified. RARβ consists of four isoforms that arise from the differential usage of two promoters and alternative splicing, which display unique expression patterns both in the embryo and in the adult. The expression of RARβ is frequently lost in many neoplastic tissues, including non-small cell lung cancer, squamous cell carcinomas of the head and neck, and breast cancer, and its expression correlates inversely with tumor grade.
