(S)-crizotinib 通过抑制MTH1来破坏核苷酸库的稳态，诱导DNA单链断裂的增加，激活人结肠癌细胞中DNA修复机制。

体内研究

在SW480 结肠癌模型中，(S)-Crizotinib (50 毫克/千克，口服，每天)损害肿瘤的生长。

激酶实验	MTH1催化测定
激酶实验	半数最大抑制浓度(IC50)通过稍微改动基于荧光的试验进行测定。简而言之，不断稀释的化合物溶于反应缓冲液中(100 mM 三羟基氨甲基醋酸盐pH 7.5，40 mM NaCl和包含0.005% 吐温-20的10 mM Mg(OAc)2 以及2 mM 二硫苏糖醇(DTT))。加入终浓度2 nM 的MTH1重组蛋白后，平板在室温条件下摇床摇动15分钟。加入底物dGTP ( 终浓度100 µM)，8-oxo-dGTP (终浓度13.2 µM)，或2-OH-dATP (终浓度8.3 µM)，MTH1对核苷酸三磷酸盐水解作用产生的焦磷酸盐(PPi)通过PPi 光无机焦磷酸盐试验试剂盒检测15分钟。用GraphPad Prism软件通过适当的剂量反应曲线对数据点做非线性回归计算IC50值。

	方法	实验前一天，细胞接种于6孔板上，培养24小时。第二天，加入DMSO(相当于化合物稀释最大量，最大值为0.2%)和逐渐增加浓度的化合物，37°C，5% CO₂条件下培养7-10天。PBS洗涤细胞后，用冰甲醇固定，结晶紫染色(0.5%的25% 甲醇溶液)，过夜晾干。对于结果的定量，将结晶紫溶于70%的乙醇，测定595nm的紫外吸收。数据用GraphPad Prism软件采用非线性回归分析。
