激酶实验 酪氨酸激酶试验
Lck HTRF激酶测定涉及胃泌素的生物素化底物肽的ATP依赖性磷酸化，在抑制剂化合物存在或不存在下进行。胃泌素的最终浓度为1.2 μM。最终ATP浓度为0.5 μM (Km app = 0.6 ± 0.1 μM)，最终LcK的浓度 (a GST-激酶域融合蛋白 (AA 225−509)) 为250 pM。缓冲液条件如下：  50 mM HEPES pH = 7.5，50 mM NaCl，20 mM MgCl2，5 mM MnCl，2 mM DTT，0.05% BSA。淬灭试验，并用160微升检测试剂停止。检测试剂为50 mM Tris，pH = 7.5，100 mM NaCl，3 mM EDTA，0.05% BSA，0.1% Tween20组成的缓冲液。阅读前，加入测试终浓度为0.0004毫克/毫升的链霉亲和素别藻蓝蛋白 (SA-APC)，以及终浓缩浓度为0.025 nM的铕抗磷酸酪氨酸Ab (Eu-抗-PY)。测定板数据在发现荧光板阅读器上，以320 nm激发光，615 和655 nm发射光进行读取。其他激酶的测试按照上诉相似的方式进行，将不同浓度的酶，多肽底物，和ATP加入反应，根据激酶的比活度，测定底物的Kms。
