| 背景 | LOX-like (LOXL)2 is a member of the lysyl oxidase (LOX) family, which is comprised of five members, the prototypical LOX and four related members, LOXL1-4. It is a secreted copper-dependent amine oxidase, and its main role is to catalyze the covalent cross-link of collagen and elastin in the extracellular matrix (ECM). LOXL2 may be a tumor suppressor and promotes invasion and metastasis in cancer. It plays a dual role in EMT induction. First, it interacts with and stabilizes Snail1, promoting CDH1 silencing and, thus, inducing EMT. Second, LOXL2 helps to maintain the mesenchymal phenotype due, at least in part, to downregulation of cell polarity and epidermal differentiation genes in a Snail1-independent fashion, associated to activation of FAK kinase. |
