| 背景 | Multidrug resistance protein 1 (MRP1/ABCC1) is a polytopic membrane transport protein that belongs to the ATP-binding cassette (ABC) superfamily and has been detected in many different drug-resistant cell lines and tumor tissues. It is capable of exporting hydrophobic drug molecules, folates, glutathione conjugated compounds, glucornoid conjugated steroids and organic anions to name a few. When overexpressed in tumor cells, MRP1 confers multidrug resistance by reducing intracellular drug concentrations in an ATP-dependent manner. In addition to the role in drug resistance, MRP1 isexpressed in non-malignant tissues and believed to be involved in protecting tissues from xenobiotic accumulation and resulting toxicity. MRP1 is expressed in most tissues throughout the body with relatively high levels found in the lung, testis, kidneys, skeletal muscle, and peripheral blood mononuclear cells while relatively low levels are found in liver. In most tissues, MRP1 is localized to the basolateral cellular surface, which in certain tissues results in the efflux of its substrates into the blood. MRP1 is expressed at very low levels in healthy human, rat, and murine liver but its basolateral expression is induced during liver regeneration, and during endotoxin and bile duct ligation-induced cholestasis. |
