背景 Ionotropic glutamate receptors (iGluRs) are a major class of heteromeric ligand-gated ion channels and mediate the majority of the excitatory neurotransmission in the vertebrate central nervous system (CNS). These transmembrane proteins can be classified as N-methyl-D-aspartate (NMDA) class or the non-NMDA class. The distribution of NMDA receptors is almost exclusive to interneurons. The N-methyl-d-aspartate (NMDA) type of ionotropic glutamate receptor is physiologically important in excitatory neurotransmission and synaptic plasticity, and pathologically important in excitatory ischaemic neural death. The rat, mouse and human genes encoding subunits of this receptor have been identified and classified into two related groups: the NMDAR1 (NR1 or ζ) subunit and the NMDAR2 subunits A, B, C and D (NR2A-D or ϵ1-4). NMDAR2A, also known as GRIN2A, regulates reward-related associative learning, cognition, memory, and structural and behavioral plasticity in the context of drug addiction. NR2A-IRP (IRP: immunoreactive protein) is enriched in cortex and hippocampus, with moderate expression in other fore-, mid- and hind-brain regions and a small amount in spinal cord. In the adult rat brain, NR2A is widespread, NR2B is mainly detected in forebrain, NR2C is concentrated in the cerebellum and NR2D is primarily found in midbrain structures and brainstem.
