| 背景 | Proprotein convertase subtilisin/kexin type-9 (PCSK9) is a secreted zymogen expressed primarily in the liver. PCSK9 circulates in plasma, binds to cell surface low-density lipoprotein (LDL) receptors, is internalized, and then targets the receptors to lysosomal degradation. Following cleavage of its signal peptide in the endoplasmic reticulum (ER), proPCSK9 is unable to leave the ER until it undergoes cotranslational autocatalytic cleavage at Gln152. In addition to low-density lipoprotein receptor, PCSK9 also has other target proteins such as VLDLR, ApoER2 and CD36. PCSK9 is regulated transcriptionally primarily by sterol regulatory element–binding protein-2 (SREBP-2), and is also regulated by hepatocyte nuclear factor-1α (HNF1α), a transcription factor that cooperates with SREBP-2 in the basal transcription of PCSK9. |
