| 背景 | The ring finger protein 8 (RNF8), a key component of protein complex, function as an E3 ubiquitin ligase that is crucial for DNA-damage response (DDR). It is ubiquitously expressed in human tissues and participates in polyubiquitination of several non-histone substrates. RNF8 mediates ubiquitin conjugation of histone at sites of damaged chromatin and also facilitates the accumulation of DDR factors through the FHA domain-mediated binding to MDC1. At DNA damage sites, RNF8 could regulate the ubiquitination of H2A, H2AX, and H2B. As a crucial component of the DDR, RNF8 exerts several pathological effects, such as neurodegeneration in neurons, radioresistance to radiotherapy in nasopharyngeal cancer, lung cancer and bladder cancer. In addition to DDR, RNF8 also contributes to regulate the rate of exit from mitosis and cytokinesis, participates in spermatogenesis, and enhances transcriptional activity of retinoid x receptor α (RXRα). |
