| 背景 | STAT1 is a cytoplasmic protein that, upon awakening from dormancy by signaling from IFNs and other factors, activates numerous genes designed, in principle, to defend the host. Activation of STAT1 occurs initially with binding of type I or type II IFNs to cognate receptors, which leads to phosphorylation-dependent activation of Janus-activated kinases (Jak1, Jak2, and Tyk2). These kinases, in turn, phosphorylate STAT1 at Tyr701. Phosphorylated STAT1 forms homodimers that are translocated to the nuclei by importin-α5 and β1 and activate the transcription of IFN stimulated genes (ISG). STAT1 forms high-affinity dimers (K d of ≈50 nM) with estimated half-lives of 20-40 min irrespective of the phosphorylation status. Unphosphorylated STATs can dimerize in an antiparallel conformation via extended interfaces of the globular N-domains, whereas STAT activation triggers a parallel dimer conformation with mutual phosphortyrosine:SH2 domain interactions, resulting in DNA-binding and nuclear retention. Tyrosine phosphorylation is dispensable for DNA binding, but the phosphorylation enforced preformed SH2 domain-mediated dimers, thus enhancing the DNA-binding activity of STAT1 >200-fold. IFN-induced STAT1 signaling primarily activates genes that mediate immune functions, antiviral and/or antipathogen functions, suppression of cell proliferation, and the induction of apoptosis. Thus, STAT1 signaling is a prodeath and tumor-suppressive pathway. |
