| 体外研究活性 | PFI-3对PB1(5)和SMARCA2/4的选择性显著高于36种其他测试的激酶[1]。PFI-3是一种有效的、具有细胞活性的蛋白相互作用抑制剂，可以选择性地结合基本的BAF溴区。在具有102种细胞受体、30种酶的筛选实验中，PFI-3只对四种GPCRs具有微摩尔级的亲和力，对BRD家族以外的蛋白没有相互作用，说明PFI-3具有良好的药物选择性。稳定性测试证明PFI3在37℃下，在液体溶液中的半衰期超过7天。在长期实验中，PFI-3能够显著地改变对干细胞分化至关重要的基因表达程序。PFI-3能够模拟Brg1缺失的效果，引起TSSs中抑制性H3K27me3标记、基因Stat3结合位点的增加[2]。PFI-3在20℃、pH7.4的PBS中，半衰期大于250 h[3]。 | |||

| 方法 | 将TSC培养于干细胞培养条件或分化培养条件下，用2 μM PFI-3或10 μM control compound处理24小时后或是整个分化诱导时期（4 days）。如果是在分化培养基中，PFI-3或control compound在诱导分化前一天开始处理。抽提RNA。 |
