    TFEB induced autophagy dependent ATZ clearance in vitro. MEF transiently co-transfected with either ATZ plus GFP expressing plasmids or ATZ plus TFEB expressing plasmids were incubated with proteasome inhibitor MG132 or with DMSO for 6 h. MG-132抑制20S 蛋白酶体ZLLL-MCA降解活性,IC50为100 nM,比ZLLal(IC50为110 μM)有效1000多倍。MG-132也会抑制钙蛋白酶,IC50 为1.2 μM。MG-132在20 nM的最佳浓度下诱导PC12细胞中神经突增生,表现出高于ZLLal 500倍的效能。[1] MG-132 (10 μM) 通过抑制蛋白酶体介导的IκBα降解,有效抑制A549细胞中TNF-α诱导的NF-κB活化,白介素-8 (IL-8)基因转录,和IL-8蛋白质释放。[2] MG-132处理通过抑制26S蛋白酶体,有效诱导p53依赖性KIM-2细胞凋亡。[3]不同于BzLLLCOCHO或PS-341,MG-132处理对26S蛋白酶体的糜蛋白样(CT-L)和肽谷氨酰基肽水解(PGPH)活性抑制较弱,而相对于BzLLLCOCHO,MG-132能够更有效诱导多发性骨髓瘤细胞(U266 和OPM-2)的细胞凋亡。[4] MG-132 (1 μM)通过激活AP-1家族成员c-Fos和 c-Jun,使TRAIL抗性前列腺癌细胞敏感,这反过来抑制抗凋亡分子c-FLIP(L)。[5] MG-132显著增强肌醇六磷酸(IP6) 的作用以降低PC3 和DU145雄激素非依赖性前列腺癌(AIPCa)细胞系的细胞代谢活性。[6]
