体内研究活性

在广泛前壁心肌梗死小鼠中,Pioglitazone通过超声心动图显著减弱左心室(LV)腔扩张和功能障碍,,以及左心室舒张末期压力。Pioglitazone部分标准化左心房 DP/ DT(最大)和DP/ DT(最小),左室收缩功能,其指标在MI小鼠中显著减少。[2]

在帕金森氏病的MPTP模型小鼠都纹状体和黑质致密部中,Pioglitazone导致小胶质细胞的激活减少,减少的感应的iNOS阳性细胞和少胶质纤维酸性蛋白阳性细胞。Pioglitazone几乎完全阻断TH阳性神经元的nitrotyrosine染色,nitrotyrosine是NO介导的细胞损伤的标记物。[3]

在帕金森氏病的MPTP模型小鼠的黑质致密部(黑质致密部)中,Pioglitazone (约20 毫克/千克/天)衰减MPTP诱导的神经胶质活化,并防止多巴胺能细胞的损失。[4]

在10个月大的APPV717I转基因小鼠的海马和皮质中,Pioglitazone导致活化的小胶质细胞和活性星形细胞数量减少。Pioglitazone处理减少了促炎的表达酶环氧合酶-2(COX-2)和诱导型一氧化氮合酶(iNOS)的表达。Pioglitazone降低beta-secretase-1 (BACE1)的mRNA和蛋白质水平,以及可溶性Abeta1-42肽的水平下降27%。[5]
