体外研究(In Vitro)
体外研究活性

ATP释放和自分泌反馈通过P2Y2和A3受体提供信号放大,控制梯度感测和中性粒细胞的迁移。[1] ATP导致活性氧(ROS)的产生,刺激磷脂酰肌醇3-激酶(PI3K)途径和随后的Akt和ERK1/2的活化。ATP依赖性ROS生成和PI3K激活还刺激氧化性应激反应所需的基因的转录。ATP介导的ROS依赖性PI3K为活化胱天蛋白酶-1以及IL-1β和IL-18的分泌所需要。[2] 在大鼠培养脑胶质细胞中,ATP有力地刺激了TNF-alpha的释放,从TNF-αmRNA表达产生。在小胶质细胞中,ATP诱导的TNF-α释放是Ca(2+)-依赖性,并且持续的Ca(2+)内流和ATP刺激的TNF-α的释放相关。ATP诱导的TNF-α释放为PD 098059所抑制,PD 098059是细胞外信号调控的蛋白激酶(ERK)激酶1(MEK1)的抑制剂,MEK1活化ERK,ATP诱导的TNF-α释放也为SB 203580所抑制,SB 203580是p38促分裂原活化蛋白激酶抑制剂。ATP在细胞外钙离子不存在的情况下迅速激活ERK和p38。[3] ATP诱导的细胞毒性是通过经典的细胞凋亡改变调控的,包括膜起泡,核缩合,DNA片段化。在小胶质细胞中,ATP但不是其它核苷酸通过P2Z受体介导的途径导致NF-κB的有效的和选择性的激活。[4]

体内研究活性

在完整小鼠大脑中,ATP调节胶质细胞分支动力学,并且从受损组织和周围的星形胶质细胞释放的ATP调控胶质细胞对损伤的快速反应。[5]
