Nocodazole是癌症相关激酶的高亲和力配体,如的磷酸化ABL, c-KIT, BRAF, 和 MEK ,Kd值分别为0.091 μM, 1.6 μM, 1.8 μM 和1.6 μM,。此外,Nocodazole对ABL(E255K)磷酸化, ABL(T315I) 磷酸化, BRAF(V600E) 和 PI3Kγ 的Kd值分别是0.12 μM, 0.17 μM, 1.1 μM 和1.5 μM。Nocodazole诱导慢性淋巴细胞白血病细胞凋亡;抑制胰岛素刺激的葡萄糖转运;在人类结肠癌细胞减少细胞凋亡;损害内侧神经节嵴细胞的形态和方向迁移[1]。Nocodazole于高浓度时,在细胞中迅速解聚微管,而低浓度可抑制微管的动态不稳定[2]。用Nocodazole处理经同步化6小时的分裂期细胞,加入不同浓度的紫杉醇后,可造成G1期阻滞,半数抑制浓度为4nM。紫杉醇处理经Nocodazole预处理的细胞,在无Nocodazole的条件下,细胞仅能形成游离微管,而在有Nocodazole存在的条件下,则可形成聚集的中心体微管[3]。 Nocodazole通过结合β-微管蛋白破坏微管,可阻碍二硫键形成和囊泡运输,抑制METH诱导的细胞凋亡和溶酶体功能障碍。Nocodazole预处理可显著减少METH诱导的细胞凋亡[4]。在iPSC中,Nocodazole可使HDR效率加倍,可高达30%[6]

Cells were treated with nocodazole (250 nM) for the length of time as indicated, and whole cell lysates were prepared. An equal amount of protein lysates was electrophoretically separated on the 10% SDS-polyacrylamide gel, and transferred to nitrocellulose membrane. Western blots were detected using specific antibodies against cyclin B1, Cdc2 (CDK1), MAD2, and Cdc20 on an enhanced chemiluminescence (ECL) apparatus. Membrane was stripped for determining the levels of GAPDH as a loading control. E (lower part). The relative protein levels for cyclin B1, Cdc2, MAD2, and Cdc20 were calculated according to their densitometry readings, which were normalized according to the corresponding readings for the GAPDH protein bands.
